When it comes to measuring protein secondary structure, there is an abundance of tools available in today’s laboratories, most commonly spectroscopy, in the form of UV Vis, Circular Dichroism (CD), or FTIR spectrophotometers. Techniques such as Raman spectroscopy are becoming more accepted with recent improvements in lasers and detectors. Even NMR has taken a step into large molecule structural analysis, with multi-dimensional NMR techniques and improved data processing.
Many of these techniques, however, come with their own limitations. CD, for instance, has a limited concentration range of about 0.1 to 0.5 mg/mL. Commonly found excipients in biopharmaceuticals such as surfactants, sugars, and amino acids are often optically active, limiting the measurement of CD on formulations. To measure a protein in its formulations conditions, Fourier-transform Infrared Spectroscopy (FTIR) may provide a solution, but not without its own challenges. FTIR is commonly a ected by background drift, making extended measurements over time challenging. Additionally, the concentration range measurable using FTIR restricts samples to greater than 10mg/mL. This limits its application to early discovery and development phases of biopharmaceutical manufacturing where such volumes are not available.
