Theme 1: Advances in Manufacturing and Analytical Characterization Working with Novel Therapeutic and Delivery Systems
Increasingly diverse and complex modalities of therapeutics and vaccines are being tested in clinics and approved for markets that go far beyond traditional small- and large-molecule platforms. These include but are not limited to mRNA and other nucleic acid-based entities, multi-specific biomolecules and novel protein constructs, combination products, multidrug conjugates, vaccines, and gene and cell therapies for rare and unmet medical needs. Complex modalities, including highly potent compounds, present complex challenges in molecule design, target product profile definition and delivery device options, manufacturing process design, controls, clinical and commercial batch release, and supply (especially under pandemic constraints). This theme aims to attract presentations on emerging technologies and predictive tools for process and product characterization as part of control strategy, critical product quality attribute assessment including physicochemical modifications/instabilities, and their biological consequences to address safety, efficacy, and regulatory concerns.
Theme 2: Challenges in Control Strategies for Drug Development
Control strategies have taken on new importance and challenges in developing and manufacturing biological and therapeutic products to control quality, efficacy, and patient safety. This theme will include developing control strategies for risk mitigations in support of pharmaceutical development and manufacturing quality, such as bioprocessing, impurities, excipients, leachables/extractables, raw materials controls, comparability studies, and expiry stability studies. Current regulatory considerations and best practices, and industry experiences for pharmaceutical product development and commercialization will be shared.
Theme 3: New Opportunities in Modeling, Controls, and Analytics
Advances in computational methodologies enable novel approaches to modeling and control with concomitant strides in product quality and process performance under accelerated timeframes. These advances are also increasingly important for next-generation therapies and to enable speedier delivery to patients. This theme will cover new opportunities in modeling, controls, and analytics to manufacture and characterize chemical and biological entities. The focus will be on the robustness of manufacturing and characterization through analytics, artificial intelligence/machine learning, advanced statistical approaches, and realtime release. Examples include strategies for model development, predictive techniques for impact of glycovariants on bioactivity/clinical performance, opportunities in continuous manufacturing, prospective controls for excipient, raw material, and ancillary materials, utilizing digital twins in design and improvement, disruptive approaches to QC/analytics, establishing expiry through modeling, updates and associated regulatory challenges, and accurate distribution and deployment of product.
